RESUMO

Hedera helix is a traditional medicinal plant. Its primary active ingredients are oleanane-type saponins, which have extensive pharmacological effects such as gastric mucosal protection, autophagy regulation actions, and antiviral properties. However, the glycosylation-modifying enzymes responsible for catalyzing oleanane-type saponin biosynthesis remain unidentified. Through transcriptome, cluster analysis, and PSPG structural domain, this study preliminarily screened four candidate UDP-glycosyltransferases (UGTs), including Unigene26859, Unigene31717, CL11391.Contig2, and CL144.Contig9. In in vitro enzymatic reactions, it has been observed that Unigene26859 (HhUGT74AG11) has the ability to facilitate the conversion of oleanolic acid, resulting in the production of oleanolic acid 28-O-glucopyranosyl ester. Moreover, HhUGT74AG11 exhibits extensive substrate hybridity and specific stereoselectivity and can transfer glycosyl donors to the C-28 site of various oleanane-type triterpenoids (hederagenin and calenduloside E) and the C-7 site of flavonoids (tectorigenin). Cluster analysis found that HhUGT74AG11 is clustered together with functionally identified genes AeUGT74AG6, CaUGT74AG2, and PgUGT74AE2, further verifying the possible reason for HhUGT74AG11 catalyzing substrate generalization. In this study, a novel glycosyltransferase, HhUGT74AG11, was characterized that plays a role in oleanane-type saponins biosynthesis in H. helix, providing a theoretical basis for the production of rare and valuable triterpenoid saponins.

In traditional Chinese medicine, Angelica dahurica is a valuable herb with numerous therapeutic applications for a range of ailments. There have not yet been any articles on the methodical assessment and choice of the best reference genes for A. dahurica gene expression studies. Real-time quantitative PCR (RT-qPCR) is widely employed as the predominant method for investigating gene expression. In order to ensure the precise determination of target gene expression outcomes in RT-qPCR analysis, it is imperative to employ stable reference genes. In this study, a total of 11 candidate reference genes including SAND family protein (SAND), polypyrimidine tract-binding protein (PTBP), glyceraldehyde-3-phosphate dehydrogenase (GAPDH), actin (ACT), TIP41-like protein (TIP41), cyclophilin 2 (CYP2), elongation factor 1 α (EF1α), ubiquitin-protein ligase 9 (UBC9), tubulin ß-6 (TUB6), thioredoxin-like protein YLS8 (YLS8), and tubulin-α (TUBA) were selected from the transcriptome of A. dahurica. Subsequently, three statistical algorithms (geNorm, NormFinder, and BestKeeper) were employed to assess the stability of their expression patterns across seven distinct stimulus treatments. The outcomes obtained from these analyses were subsequently amalgamated into a comprehensive ranking using RefFinder. Additionally, one target gene, phenylalanine ammonia-lyase (PAL), was used to confirm the effectiveness of the selected reference genes. According to the findings of this study, the two most stable reference genes for normalizing the expression of genes in A. dahurica are TIP41 and UBC9. Overall, our research has determined the appropriate reference genes for RT-qPCR in A. dahurica and provides a crucial foundation for gene screening and identifying genes associated with the biosynthesis of active ingredients in A. dahurica.

RESUMO

Air quality is a significant environmental issue among the Chinese people and even the global population, and it affects both human health and the Earth's long-term sustainability. In this study, we proposed a multiperspective, high-dimensional spatiotemporal data visualization and interactive analysis method, and we studied and analyzed the relationship between the air quality and several influencing factors, including meteorology, population, and economics. Six visualization methods were integrated in this study, each specifically designed and improved for visualization analysis purposes. To reveal the spatiotemporal distribution and potential impact of the air quality, we designed a comprehensive coupled visual interactive analysis approach visually express both high-dimensional and spatiotemporal attributes, reveal the overall situation and explain the relationship between attributes. We clarified the current spatiotemporal distribution, development trends, and influencing factors of the air quality in China through interactive visual analysis of a 25-dimensional dataset involving 31 Chinese provinces. We also verified the correctness and effectiveness of relevant policies and demonstrated the advantages of our method.

In order to investigate and assess the distribution of pathalic acid easters (PAEs) in agricultural products from typical areas of the Pearl River Delta, South China, 131 agricultural products were sampled for determination of 6 PAEs priority pollutants classified by the U. S. EPA by GC-FID. The results showed that the total contents of the PAEs (sigma PAEs) in agricultural products samples ranged from nd to 79.86 mg x kg(-1) and the mean value was 2.84 mg x kg(-1), with the detected ratio of 98.5%. The average concentrations of sigma PAEs in different types of agricultural products were ordered by vegetables (3.03 mg x kg(-1)) > rice (2.52 mg x kg(-1)) > fruits (1.26 mg x kg(-1)). The mean concentration of PAEs distributed in the four typical cities of the Pearl River Delta, and decreased in the sequence of Zhuhai (6.53 mg x kg(-1)) > Dongguan (2.59 mg x kg(-1)) > Huizhou (1.53 mg x kg(-1)) > Zhongshan (1.12 mg x kg(-1)). Di-n-butyl phthalate (DBP), di-(2-ethylhexyl) phthalate (DEHP) and di-n-octyl phthalate (DnOP) contributed more than 90. 8% of the total PAEs in samples, and were the main components of PAEs in agricultural products from the Pearl River Delta, with higher percentage contents and detected ratio. Meanwhile, the average concentrations of sigma PAEs in cabbage mustard, lettuce occurred in Zhuhai and Dongguan cities, followed by lettuce and leaf lettuce in the corresponding DEHP from Zhuhai city, both exceeded the suggested standards in U.S.A. and Europe and were of high health risk. There were significant differences among 14 various vegetables in the contents of the 6 PAEs compounds, and the sigma PAEs contents in cabbage mustard and lettuce as part of leafy vegetables were higher than those in other vegetables, while the lowest were detected in flowering cabbage and edible amaranth. Therefore, the type of vegetables and its growing environment exposed to the atmosphere and soil were the main factors that significantly affected their accumulation of PAEs concentrations.

Cross-cultural psychologists assume that core cultural values define to a large extent what a culture is. Typically, core values are identified through an actual self-importance approach, in which core values are those that members of the culture as a group strongly endorse. In this article, the authors propose a perceived cultural importance approach to identifying core values, in which core values are values that members of the culture as a group generally believe to be important in the culture. In 5 studies, the authors examine the utility of the perceived cultural importance approach. Results consistently showed that, compared with values of high actual self-importance, values of high perceived cultural importance play a more important role in cultural identification. These findings have important implications for conceptualizing and measuring cultures.
